Raji cells (1 × 105 cells per well of a 96-well plate) were incubated for 1 hour at room temperature with either recombinant rituximab, sera from mice injected with HEDGES vector encoding rituximab and lipid, or control mouse sera (mice injected with hG-CSF and lipid). Twenty microliters of pooled normal human plasma (from Innovative Research) was then added to all wells, and the plates were incubated for another 12 hours at 37°C. Samples were run in duplicate. Cell viability was measured using the Promega CellTiter-Glo reagent according to the manufacturer’s instruction (G7570).
