Jurkat CD4+ T cells were infected by incubation of 1 million cells with HIV-1 NL4.3 Gag-iGFP particles in 50 μl of RPMI medium for 1 hour at 37°C. Cells were washed three times in RPMI medium and then cultured for 72 hours at 2 million cells per milliliter to achieve a 5 to 10% infection rate with progeny virus production.
