Plasmid and poly(I:C) transfections were performed using minor modifications to the manufacturer’s protocol. One hour before transfection, fresh medium was added to cells. Plasmid or High Molecular Weight poly(I:C) (tlrl-pic, InvivoGen) and Lipofectamine 2000 (11668027, Thermo Fisher Scientific) were diluted separately into Opti-MEM, vortexed briefly, and incubated at room temperature for 5 min. The two solutions were mixed, vortexed briefly, and incubated at room temperature for 20 min. Complexes were added dropwise to cells and placed in the incubator for 5 hours. Medium was then replaced with medium containing the indicated compounds.
