WT E. coli and CobB KO E. coli strains were cultured in M9 minimal medium with 0.2% glucose and l-lysine (100 mg/ml) (CobB KO) or 13C6-lysine (WT), respectively. Then, cells were harvested during the mid-exponential phase of growth and lysate, and HPLC fraction and HPLC-MS/MS analysis were prepared for acylated peptide enrichment as described above.
