Ninety-six–well plates were coated with either fibronectin or collagen I for an hour at 37°C, followed by an hour of incubation with 0.5% BSA-containing blocking buffer. HEK293 or MCF10A cells were seeded at a density of 4 × 105, and cells were allowed to attach for 45 min at 37°C. The cells were washed three times with 0.1% BSA-containing culture media, and then, the cells were fixed with 4% PFA. Next, the cells were stained with crystal violet for 10 min. After staining, the cells were washed 10 times with distilled water. Cells were air-dried for an hour and solubilized with 2% SDS on an agitator. The absorbance of each well was measured at 550 nm to quantify the adhesion properties of cells.
