The concentrations of total IgG in serum from asthmatic mouse model were evaluated with enzyme-linked immunosorbent assay kits according to the manufacturer’s instructions (BioLegend). For measurement of Ab to LCMV, purified LCMV antigen (0.5 μg/ml) was coated onto a 96-well Costar assay plate (Corning). Plates were blocked with 1% bovine serum albumin (BSA) in phosphate-buffered saline (PBS), and serial dilutions of sera in 1% BSA in PBS were applied to the plates and incubated at room temperature for 2 hours. After washing, plates were incubated for 2 hours with the horseradish peroxidase–conjugated Ab against total mouse IgG (Jackson ImmunoResearch) and then with 3,3′,5,5′-tetramethylbenzidine substrate (BioLegend) for color development. Absorbance was measured at 450 nm with a microplate reader (Molecular Devices).
