The green curves in Fig. 3B were obtained computationally by simulating I0 and I1 for isolated point emitters in MATLAB. First, the intensity distribution of a point source imaged with the fiber bundle using the large aperture setting (tan θ1 = 0.4534, σ0 = 2.30 μm) was calculated using the assumed PSF in Eq. 3. This calculation was performed for point emitters at depths z = 1, 11, …, 101 μm and then repeated for a small-aperture setting (tan θ0 = 0.3574, σ0 = 2.30 μm). This yielded simulated images I1 and I0 for each bead depth. Light field calculation was then performed exactly as it is in the case with experimental data (see “Effective light field moment calculation” section).
