Magnetic affinity cell sorting was done using commercially available EasySep kits (CD14, CD19, CD4, and CD8 positive isolation II and T cell negative isolation) from STEMCELL Technologies. To implement the isolation protocols on the device, the buffers were modified and volumes were scaled accordingly. EasySep buffer (STEMCELL Technologies) was supplemented with 10% FBS (Gibco) and 0.2% Pluronic-F127 (Sigma), to reduce nonspecific cell adhesion in the polydimethylsiloxane channels. The microchannels were also preincubated with 1% Pluronic-F127 (Sigma) before cell isolation. Neodynium magnets (Grainger) with a 43-lb pull were used for all magnetic capture steps.
