Cy5-NBD-PE-cRGDLs or Cy5-NBD-PE-PLs (equivalent to 26.5 mg SPC/kg) were administrated to I/R rats induced by the reperfusion of the MCAO. Blood of 200 μl was collected at the desired time points after reperfusion, treated with 2 ml of red cell lysates for 5 min of incubation, and subsequently centrifuge at 3300 rpm. The supernatant was discarded, and the precipitate was resuspended for the collection of leukocytes. The percentage of dual-positive (Cy5 and NBD-PE) M/Ns in leukocytes were assayed by flow cytometer (Fig. 2A). Blood (1 ml) was collected at the end of the experiment for the isolation of neutrophils/monocytes using the density gradient centrifugation method and visualized by CSLM (Fig. 2B).
