Medium was collected after 24 hours of culture from two hypoxic and four nonhypoxic hydrogels per experiment to normalize for cell number. Immediately after collection, protease inhibitors (Halt Protease and phosphatase inhibitor cocktail) were added to the medium, and then the medium was stored at −80°C until use. Arrays were prepared according to the manufacturer’s protocol. Films were exposed for 20 min and then analyzed using ImageJ (public domain) and graphed using GraphPad Prism 6 (GraphPad Software Inc.). Values of mean pixel density were reported.
