We created average gene expression profiles (TPM) for each individual cell types. To identify cell type–specific TFs, we used a list of known TFs described earlier (35) and crossed it with TPM-normalized expression by cell types. Only TFs with expression greater than or equal to 0.75 TPM and expressed in more than 10% of cells were considered. For each selected TF, a Z score based on log2(TPM + 1) was calculated, and to find TFs highly specific to an individual cell type, we applied a threshold of 0.6 to the ratio of second maximal expression value to the maximal. For the graphical representation of results, we selected top TFs (maximum of 10) with the highest Z score for each cell type.
