Ultrafiltration for enrichment and buffer exchange

All samples (cross-linked and uncross-linked) were subjected to three rounds of ultrafiltration (Amicon Ultra 500 μl with 100k cutoff). Ultrafiltration was carried out at 20°C and 7k relative centrifugal force (Eppendorf 5424R). The buffer was replaced by folding buffer (5 mM tris, 1 mM EDTA, and 5 mM NaCl; including 5 mM MgCl2), PBS, or double-distilled water supplemented with 300, 150, 100, 50, 25, and 0 mM NaCl. Samples used for cryo-EM were concentrated to 1000 nM.
