摘要:酵母细胞表面展示是一种固定化表达异源蛋白的真核展示系统,在文库筛选、生物吸附、免疫检测和生物传感器研发等领域发挥重要作用。外源蛋白通过酿酒酵母a凝集素展示在细胞表面。a凝集素包含核心亚基AGA1及结合亚基AGA2。AGA1与酵母细胞壁β-葡聚糖共价连接,并通过两个二硫键与AGA2的N端共价相连。外源蛋白与AGA2的C端融合,从而实现外源蛋白在酿酒酵母细胞的表面展示。本实验通过AGA1和AGA2将外源蛋白固定在酵母细胞表面,获得菌酶一体化的微生物细胞反应器。
