摘要：Northern blot是指在变性条件下，将待测的RNA进行琼脂糖凝胶电泳分离，继而将凝胶中的样本转移到硝酸纤维素膜或者尼龙膜，固定后再与地高辛 (Digoxigenin) 标记的RNA探针进行杂交反应，最后根据地高辛标记情况分析杂交结果的分子生物学技术。Northern blot 主要用于检测目的基因的存在，不同长度的转录本及相对含量。对于同位素标记，Digoxigenin标记的Northern blot具有环境友好和更加方便操作的优势。

实验原理

在变性条件下将待检测的RNA样品进行琼脂糖凝胶电泳，继而将在凝胶中的位置转移到硝酸纤维素薄膜或尼龙膜上，固定后再与地高辛或其它标记物标记的RNA探针进行杂交检测 (图1)。
