研究背景

在秀丽隐杆线虫的RNA干扰过程中，外源的双链RNA（dsRNA）被注射或喂食到线虫体内后，被DICER切割成短的dsRNA，再由RDE-1结合，进而诱导靶标mRNA的降解沉默。之后在RdRP（RNA dependent RNA Polymerase，RNA依赖的RNA聚合酶）的作用下，诱导次级siRNA，即22 GRNA生成，并被WAGO蛋白结合。另外，线虫体内还存在有内源性的初级siRNA，这类小RNA长为26nt，5’末端为G且有单磷酸修饰，被称为26G RNA。26G RNA也可以作为沉默的初级信号，诱导次级小RNA，22G RNA的生成(Billi et al., 2014)。22G RNA长为22 nt，5’末端为G且具有三磷酸修饰，使得该RNA并不能直接建库测序。对这一类22G RNA进行测序时，需要将RNA的5’末端的三磷酸处理为单磷酸结构，进而进行建库测序(Ruby et al., 2006; Gu et al., 2012)。其中CIAP可以将RNA 5’末端的磷酸基团去除，转为羟基，而T4 PNK可以将5’末端羟基加上单磷酸修饰。使用CIAP和T4 PNK处理，可以对三磷酸修饰的小RNA后续的建库和测序。
