摘要：造血祖细胞 (Hematopoietic Progenitor Cells, HPCs)是由造血干细胞分化来的，具有定向分化潜能的造血前体细胞。造血祖细胞主要分为4类：common myeloid progenitors (CMPs)，common lymphoid progenitors (CLPs)，granulocyte-macrophage progenitors (GMPs) 和megakaryocyte-erythrocyte progenitors (MEPs)。分析造血祖细胞可研究造血干细胞向各系分化的能力。本文以小鼠骨髓组织为例，使用7色共14种抗体，对4类主要的造血祖细胞 (CMPs、CLPs、GMPs和MEPs) 进行标记。

材料与试剂

  1. 6孔板
  2. 15 ml离心管
  3. 1.5 ml EP管
  4. 70 μm滤膜 (Fisherbrand, catalog number: 22363548)
  5. 小鼠 (C57BL/6J，6~8周龄，雄雌各半)
  6. 胎牛血清 (FBS，Gibco，catalog number: 10100147)
  7. 流式抗体 (见表1）
  8. 同型对照及单标抗体 (见表 2）
  9. 磷酸缓冲液 (10x PBS, Sangon biotech, catalog number: E607016-0500)
  10. 氯化铵 (天津大茂化学，catalog number: 12125-02-9)
  11. 碳酸氢钠 (Sigma-Aldrich, catalog number: S5761，500 G)
  12. EDTA粉末 (鼎国，catalog number: LE568)
  13. 磷酸缓冲液 (1x PBS) (见溶液配方)
  14. 红细胞裂解液 (500 ml) (见溶液配方)
