Larval imaginal discs were dissected out and washed briefly in cold PBS. Discs were kept in Acridine orange (AO) for 2–3 min followed by two gentle washes in PBS. Then, the discs were mounted and observed under a Nikon Eclipse 80i fluorescence microscope (Nikon, Garden City, NY).
